Decoding the Impact of RGL3 Genetic Variants on Protein Structure and Binding Site: A Bioinformatics Approach to Uncover Potential Hypertension Associations

Background: The RGL3 gene plays a role in key signal transduction pathways and has been implicated in hypertension risk through the identification of a copy number variant deletion in exon 6. Genome-wide association studies have highlighted RGL3 as associated with hypertension, providing insights into the genetic underpinnings of the condition and its protective effects on cardiovascular health. Despite these findings, there is a lack of data that confirms the precise role of RGL3 in hypertension. Additionally, the functional impact of certain variants, particularly those classified as variants of uncertain significance, remains poorly understood. Objectives: This study aims to analyze alterations in the RGL3 protein structure caused by mutations and validate the location of the ligand binding sites. Methods: Clinical variants of the RGL3 gene were obtained from NCBI ClinVar. Variants of uncertain significance and likely benign were analyzed. Multiple sequence alignment was conducted using BioEdit v7.7.1. AlphaFold 2 predicted the wild-type and mutant 3D structures, followed by quality assessment via PROCHECK. Functional domain analysis of RasGEF, RASGEF_NTER, and RA domains was performed, and BIOVIA Discovery Studio Visualizer 2024 was used to evaluate structural and physicochemical changes. Results: The analysis of 81 RGL3 variants identified 5 likely benign and 76 variants of uncertain significance (VUS), all of which were missense mutations. Structural modeling using AlphaFold 2 revealed three key domains: RasGEF_NTER, RasGEF, and RA, where mutations induced conformational changes. Ramachandran plot validation confirmed 79.7% of residues in favored regions, indicating an overall reliable structure. Moreover, mutations within RasGEF and RA domains altered polarity, charge, and stability, suggesting potential functional disruptions. These findings provide insight into the structural consequences of RGL3 mutations, contributing to further functional assessments. Discussion & Conclusion: The identified RGL3 mutations induced physicochemical alterations in key domains, affecting charge, polarity, hydrophobicity, and flexibility. These changes likely disrupt interactions with Ras-like GTPases, impairing GDP-GTP exchange and cellular signaling. Structural analysis highlighted mutations in RasGEF and RA domains that may interfere with activation states, potentially affecting protein function and stability. These findings suggest that mutations in RGL3 could have functional consequences, emphasizing the need for further molecular and functional studies to explore their pathogenic potential.

Detection of salivary biomarker for migraine diagnosis

Detection of salivary biomarker for migraine diagnosis

Migraine, a prevalent neurological disorder, is the third most common disease globally, causing significant health and financial burdens. It has four stages: prodrome, aura, headache, and postdrome. The prodrome (also known as premonitory) stage is crucial as it precedes the headache by up to 72 hours. Taking medication during the premonitory peroid has shown to prevent the headache phase . However, the symptoms of premonitory period lack specificity, making it difficult for patients to know if they’re experiencing premonitory symptoms. Calcitonin-gene related peptide (cGRP),is a protein that plays a key role in migraine pathogenesis and studies found that salivary cGRP levels increase during the premonitory stage. This study aims to develop and evaluate a lateral flow immunoassay kit for detecting salivary cGRP levels in migraine patients during the prodrome stage. It can serve as a confirmation tool for premonitory symptoms.

Application for skincare analysis and suggestion

Nowadays, consumers may have experienced situations where they don't know which product to use based on their skin problems, the product they use doesn't give the desired results, the product is not worth the price, allergic to certain chemicals in the product ,or use multiple products and have ingredients that should not be used together leading to irritation. For this reason, we develop an application to analyze skin care product ingredients to solve such problems. This allows consumers to correctly understand information about the ingredients in products and know which products they should use according to their skin problems without having to rely on chemical knowledge and get products that are the best value for money. The project has integrated software knowledge to develop an application for analyzing skin care product ingredients. To find and recommend suitable skin care products to consumers. The information on various important ingredients has been collected from reliable articles and research.

Smart Jaundice Diagnostic System

Jaundice, a common condition in infants that results from high bilirubin levels in the blood, often requires early diagnosis and monitoring to prevent severe complications, especially in newborns. Traditional diagnostic methods can be time-consuming and subject to human error. This study proposes an approach for real-time jaundice detection using advanced image processing techniques and machine learning algorithms. By analyzing images captured in RGB color spaces, pixel values are extracted and processed through Otsu’s thresholding and morphological operations to detect color patterns indicative of jaundice. A classifier model is then trained to distinguish between normal and jaundiced conditions, offering an automated, accurate, and efficient diagnostic tool. The system’s potential to operate in real-time makes it particularly suited for clinical settings, providing healthcare professionals with timely insights to improve patient outcomes. The proposed method represents a significant innovation in healthcare, combining artificial intelligence and medical imaging to enhance the early detection and management of jaundice, reducing reliance on manual interventions and improving overall healthcare delivery.

Investigation of Microalgae Chorella sp. KLSc61 crude extract to promote Lactobacillus plantarum JCM 1149 Growth

Microalgae are rich in bioactive compounds that may contribute to the growth of probiotics, which require appropriate nutrients, known as prebiotics, to thrive. This study aims to evaluate the effectiveness of crude extracts from intracellular components residues of the microalga Chlorella sp. KLSc61 in promoting the growth of the probiotic bacterium Lactiplantibacillus plantarum JCM1149 under simulated gastrointestinal conditions. The intracellular extracts were obtained using 70% (v/v) ethanol, and their effects on probiotic growth were tested at concentrations of 0.1%, 0.75% and 1.5%. The growth of Lactiplantibacillus plantarum JCM1149 was assessed using the drop plate method. The findings of this study will provide insights into the potential of Chlorella sp. KLSc61 extracts in enhancing probiotic growth, which could lead to the development of synbiotic dietary supplements containing both probiotics and prebiotics. Additionally, this study may serve as a foundation for further research on the role of microalgal extracts in gut health and immune system modulation.
